Apparently, there were some changes in the libpng version that comes with Lion, so the current (Snow Leopard based) version of Matplotlib won't work on Lion. Hopefully this will be fixed in an upcoming binary installer for Matplotlib but until then you'll have to use the homebrew package manager (see link).
